Captain Cian Prendergast's fourth-minute try was the catalyst for a tenacious Connacht performance at Scotstoun Stadium, which saw them stay in contention until the closing stages.
A second-half brace from player-of-the-match Kyle Steyn, with his last effort six minutes from the end, was crucial in guiding the table toppers through to a home semi-final against Munster or the Vodacom Bulls.
Stuart Lancaster's men kept them scoreless during Sam Illo's sin-binning, although an earlier Patrick Schickerling try meant it was seven points apiece at half-time.
Steyn and Josh McKay gave Glasgow a 21-7 cushion, only for Dave Heffernan and Finlay Bealham to strike back - either side of a Jare Oguntibeju score - as Connacht twice made it a seven-point game.
Injuries hampered the visitors, though, with Shayne Bolton, Hugh Gavin, and Bealham all having to come off, and Steyn crossed for a second time to end their hopes.
Connacht can still take a huge amount from their season run-in, which saw them win eight of their last nine URC matches for an eighth place finish and qualification for next season's Investec Champions Cup.
The province made an impressive start to their first URC play-off appearance since 2023. The impressive Josh Ioane and Bolton both made ground with ball in hand.
They had the Glasgow defence under early pressure, being held up from one prolonged attack before Prendergast drove over with Darragh Murray on the latch.
Following Sam Gilbert's first conversion of the night, Glasgow used a couple of penalties to get within striking distance, and a third infringement saw Connacht receive a team warning.
Another bout of forward pressure ended with Namibian-born prop Schickerling scrambling over from a pick-and-go. George Horne added a levelling conversion from the left.
The Scots swiftly came hunting again, as incisive running from Horne and Sione Tuipulotu had the visitors on the retreat, and Illo saw yellow for offside in the 14th minute.
Glasgow had a try disallowed during the prop's absence, as Matt Fagerson was penalised for obstruction from a restarted maul that saw Johnny Matthews touch down.
While the Warriors continued to have the better of the exchanges, 14-man Connacht regained lost ground through pacy half-backs Ben Murphy and Ioane, and the bustling Bolton.
Although they could not capitalise on an Ioane 50:22 kick, Illo returned with the sides still level at seven-all, and the pace of this entertaining encounter frenetic at times.
Picking up a loose Glasgow pass, New Zealander Ioane had his sights set on the left corner but Steyn reeled him in with a timely try-saving intervention.
The pressure was back on the Connacht defence at the start of the second half. A Bundee Aki turnover helped to keep Glasgow at bay.
That was until five minutes in when Steyn, armed with a penalty advantage and following on from his own 50:22 kick, sliced through a gap to make it 14-7.
Matt Fagerson missed out on a try soon after, due to a forward pass from Dempsey. Bolton had hauled down the fast-breaking Horne before that, but injured himself in the process.
Connacht turned to their bench with a new front row introduced, and Seán Naughton brought on out wide. Rory Darge and Peter Dooley exchanged turnovers as the breakdown battle intensified.
The combination of clever tactical kicking and a couple of scrum penalties paved the way for McKay to dart over in the 55th minute, via an inviting Dan Lancaster pass.
Prendergast led Connacht's response, ensuring a counter attack breached the opposition 22. Disappointingly, they lost the influential Ioane to a head injury following a collision with Alex Samuel.
Nonetheless, three penalties in quick succession had the westerners chasing their second try, which arrived in the 64th minute when Heffernan burrowed over with support from Joe Joyce.
The close-range score was awarded after a TMO review, and with Glasgow also losing lock Samuel to the sin bin, Gilbert's conversion left seven points in it again at 21-14.
Frustratingly for Connacht, there was more disruption to their back-line as Gavin was stretchered off with a serious leg injury, and Glasgow soon hit back with an opportunist fourth try.
South African replacement Oguntibeju picked from a ruck, outside Connacht's 22, to break for the try-line and crash over, allowing out-half Lancaster to make it a double-scores lead.
Matthew Devine was the spark for Connacht, his scampering break preceding some strong carrying from the pack. Bealham timed his pick perfectly, backed up by Heffernan, for a 72nd-minute grounding.
Lively replacement Seán O'Brien was prominent in the build-up, but despite Gilbert's right boot shaving the deficit down to seven points once more, Steyn had the final say.
After O'Brien misjudged the restart, conceding a five-metre lineout, Glasgow got Lancaster in position to fire out a skip pass for Steyn to raid over out wide.
With Bealham unable to continue, Connacht ended the game down to 14 men as Illo had to come back on to ensure contested scrums, and Shamus Hurley-Langton was the player to make way.
