Following back-to-back unbeaten seasons, this was a first defeat for Leinster at this age-grade since 2023, as Munster got their campaign off to a confidence-boosting start at Virgin Media Park.
Second-half tries from Charlotte Deegan and Cora Miller, coupled with a brace of penalties from Richmond-Scariff starlet Eden Waters, has the Reds out of reach.
It had been 10-7 to the visitors at half-time, with Mary O'Hara's 28th-minute try, whilst Leinster had Sophie McDonagh in the sin bin, cancelling out a converted Caitríona Conroy effort.
Sophia White opened the scoring for the title holders with a 19th-minute penalty, yet Munster proved to be clinical during the final half-and-hour, scoring 20 points in as many minutes.
There were three yellow cards after the break - two for Damien Hicks' home side - and replacement Ruby Osumah's last-gasp try gave Leinster a positive finish, ahead of their only home match next Saturday against Connacht.
The second round will see Munster return to their Cork base to face Ulster, hoping to take another step towards earning a place in the August 30 final at Mullingar RFC.
The breakdown was hotly contested right from the off as the sun shone brightly on Leeside. Sadhbh Brennan secured an early penalty for the hosts as the Charlie Doel-coached Leinster team probed for openings.
Arklow out-half White, a central figure for the Blues, engineered the game's first break before Lily Harney's attempted pass back to her - as space opened up - went forward.
Munster advanced downfield thanks to a series of penalties, including one won by Waters at the breakdown. However, captain Emily Miller led Leinster's impressive maul defence.
Despite a well-won turnover by Brennan, Leinster number 8 Emma O'Rourke threatened the try-line soon after, following up on O'Hara's pacy run into the Munster 22.
Munster's defence was resilience - their forwards doubling up in tackles, and Waters got another breakdown decision - but a subsequent penalty allowed White to curl over the opening points.
Hicks' charges responded by getting their back-three into attacking situations, while Waters used a clever dummy and stayed on her feet to bring them within five metres of the whitewash.
Successive offsides landed Leinster centre McDonagh in the sin bin. Alice Buckley went close from the resulting tap penalty, before Dungarvan flanker Conroy swiftly picked to score, with Waters also converting.
Nonetheless, the visitors hit back within three minutes. White went quickly from a penalty and provided the pass, following good interplay between Harney and O'Rourke, for O'Hara to cross from close range.
The teams both exerted pressure either side of half-time, with Leinster unable to take advantage of Caoimhe O'Mahony's interception and a late lineout platform, while Munster began the second half very much on the front foot.
An increasing amount of knock-ons made for a scrappy spell, and although Leinster skipper Miller's incisive run made almost 20 metres, White nudged a penalty attempt narrowly wide.
Still, Doel's charges were quickest to react to the missed place-kick, swooping to retrieve possession. O'Hara would have doubled her try tally but for a forward pass from Harney.
It remained 10-7 to Leinster until Waters punished a penalty, conceded by White for blocking, with a fine touchfinder. Fethard prop Deegan then did brilliantly to bulldoze her way over from the base of a ruck.
Waters coolly converted to make it 14-10, and superbly split the posts from near the left touchline after Cora Miller had added a quick-fire 51st-minute score.
The Waterpark winger had the pace to capitalise on a loose ball that came from an initial Waters kick, racing over from 30 metres out. Suddenly, Leinster were staring at an 11-point deficit.
They had some joy down the left hand side with Portlaoise flyer O'Hara twice breaching the Munster 22, and too many team penalties resulted in Deegan seeing yellow.
However, Danielle Naughton won a crucial turnover, allowing Munster to move back downfield. Sprung from the bench, Sadhbh Whelan carried strongly, and Waters turned a 63rd-minute penalty into an important three-pointer.
Munster regained good field position from the restart, with Leinster replacement Síofra McGlynn sin-binned for a high tackle. Possession changed hands, but Whelan forced a breakdown penalty and Waters delivered from distance for a 27-10 scoreline.
Leinster lifted the tempo in response, getting their big ball carriers involved as Munster lost Róisín Cullinane to the bin. The powerful Osumah duly drove over, following a tap penalty during the final play.
